| PTEN
Phosphatase and tensin homolog (PTEN) is a phosphatase and is encoded by the PTEN gene.Mutations of this gene are a step in the development of many cancers, specifically glioblastoma, lung cancer, breast cancer, and prostate cancer.PTEN protein acts as a phosphatase to dephosphorylate phosphatidylinositol (3,4,5)-trisphosphate (PtdIns (3,4,5)P3 or PIP3).PTEN specifically catalyses the dephosphorylation of the 3` phosphate of the inositol ring in PIP3, resulting in the biphosphate product PIP2 (PtdIns(4,5)P2). This dephosphorylation is important because it results in inhibition of the Akt signaling pathway, which plays an important role in regulating cellular behaviors such as cell growth, survival, and migration.
| Repeatability
Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%| Recovery
The recovery of Mouse PTEN spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.| Sample Type | Range(%) | Average Recovery(%) |
| Serum(n=8) | 84-101 | 96 |
| Plasma(n=8) | 92-106 | 102 |
| Cell Culture Supernates(n=8) | 96-108 | 107 |
| Linearity
Samples were spiked with high concentrations of Mouse PTEN and diluted with Universal Diluent to produce samples with values within the range of the assay.| Serum(n=4) | Plasma(n=4) | Cell Culture Supernates(n=4) | ||
| 1 : 2 | Range (%) | 84-95 | 88-96 | 90-110 |
| Average (%) | 91 | 93 | 96 | |
